TITLE: GPCR heterodimerization leads to opioid-induced hyperalgesia.

ABSTRACT: Opioids are the most prescribed and effective drugs for treatment of moderate and severe pain. Despite their pharmacological efficacy, the use of opioids in therapy is compromised by the onset of significant side effects as opioid-induced hyperalgesia. We have recently discovered a new isoform of μ-opioid receptor, which consists of six trans-membrane helices (6TM-mOR) that is able to dimerize with β2 adrenergic receptor (b2AR). Upon migration on the cellular surface, the 6TM/b2AR heterodimer is activated by morphine and leads to the development of severe opioid-induced hyperalgesia. Such finding can drive through the therapeutic use of classical beta-blockers in the treatment of chronic pain.

ABSTRACT: In recent years two computational intensive approaches evolved; At the molecular level, in-silico rational drug design methods have been developed to suggest novel chemotherapeutic agents. The methods include compound library design, homology modeling, binding site prediction, etc. Binding affinity prediction is being performed with docking techniques, molecular dynamics, and hybrid quantum mechanics/molecular mechanics approaches. At a higher level, the systems biology approach aims at modeling the highly interconnected nonlinear characteristics of biological systems. These efforts span many types of intra- and inter- cellular networks. I order to design and test in-silico new chemotherapeutic treatments, a fusion of both methods is required, allowing bottom-up and top-down feedback.
